Isopeptidase T is a member of the high molecular weight ubiquitin C-terminal hydrolases (UCHs) that are a family of cysteine hydrolases that catalyze the hydrolysis of amides, esters and thioesters from the C-terminus of ubiquitin. This enzyme primarily disassembles free polyubiquitin chains linked through isopeptide bonds. This activity has a critical role in the recycling of Ub and in the regulation of the activity of the 26S proteasome. This isoform results from a splicing variant of the long isoform, and differs by a substitution of 23 amino acids (G629-S652) with a single alanine.
