Angiopoietin-2 (ANGPT2) is a ligand of the receptor tyrosine kinase TIE2 (TEK). In conjunction with vascular endothelial growth factor (VEGF), ANGPT2-TIE2 signaling induces vascular sprouting. Inhibition of ANGPT2 shows promising results in preventing tumor angiogenesis, tumor growth, and metastasis in preclinical models, making it an attractive candidate to target in cancer.

Angiopoietins are a family of secreted factors consisting of ANGPT1, ANGPT2, ANGPT3 (ANGPTL1), ANGPT4, ANGPT5 (ANGPTL3), ANGPTL2, ANGPTL4, ANGPTL5, ANGPTL6, and ANGPTL7. ANGPT1 is a potent TIE2 activator, whereas ANGPT2 functions as a context-dependent agonist/antagonist. ANGPT2 is highly upregulated at the early stages of tumor progression to drive neovessel formation. Conventional anti-VEGF therapies primarily target immature blood vessels in tumors. The combinational targeting of both ANGPT2 and VEGF pathways is now expected to improve clinical efficacy by expanding the target vessel population.
