NEW STYRENE-MALEIC ANHYDRIDE (SMA) POLYMERS

 

What are they?

  • Four, novel, modified SMA polymers, designed to form planar lipid bilayer nanodiscs [1]
  • Direct, detergent-free systems for the extraction of membrane proteins (MPs) in their natural membrane environment
  • Used for the reconstitution of MPs into synthetic lipid nanodiscs
  • Demonstrated to solubilize well-folded MPs [1]
  • Good for structural and functional studies of MPs

Why use them?

  • Increased stability in presence of divalent cations: up to 200 mM Ca2+ or Mg2+ (see Fact Sheets or [2])
  • These new molecules cover the full range of pH stabilities [2]
  • Easy nanodisc size control: adjust by altering the polymer:lipid ratio
  • Capable of reconstituting MPs with large, charged, soluble domains [1]
  • Easy nanodisc formation protocol: add directly to membranes or lipids, incubate, then proceed to purification or other techniques
  • Compatible with most protein purification and characterization techniques

  • Studies of membrane proteins in native lipid environments
  • Structural studies of large membrane proteins and complexes
  • Studies of protein:lipid interactions
  • Work with proteins that are unstable in detergents
