How to choose a tag antibody?
A protein tag is a peptide or protein that is expressed in fusion with a target protein using in vitro recombinant DNA technology to facilitate the expression, detection, tracing and purification of the target protein.
Tag antibodies can be used to detect the expression, intracellular localization and purification of tag fusion proteins, as well as qualitative or quantitative detection of tag fusion proteins.
So how should we choose labeling antibodies in our experiments?
|
His Tag
Consists of six consecutive histidines and is commonly used for protein purification by metal ion affinity chromatography (IMAC). Advantages include mild purification conditions, small tags and low impact on protein function.
|
|
GST Tag
Glutathione S-transferase, advantages include enhanced solubility of exogenous proteins, maintenance of protein antigenicity and biological activity, but higher molecular weight may affect protein function.

MBP Tag
Maltose-binding protein that contributes to increased proteolysis, especially for eukaryotic proteins. Can be purified by starch affinity chromatography, but larger tags may have an effect on protein structure/function.
