DNA-encoded Library (DEL) |
DNA-encoded library (DEL) is a technology where each small molecule compound is linked to a unique DNA sequence at the DNA level, enabling DNA encoding similar to assigning a “barcode” to each molecule.
DEL screening is a technique used in the early stages of drug discovery to rapidly screen billions of small molecule compounds in order to identify new chemical structures for further validation and optimization. |
Workflow of Solid-phase DEL Screening |
1. |
The target protein (blue), immobilized on solid support, is incubated with DEL. |
2. |
Unbound library members are washed away by means of consecutive washing steps. |
3. |
Library members bound to the target protein are eluted. |
4. |
After final elution, the DNA tags on the eluted protein-binding library members are amplified by PCR. |
5. |
The amplified DNA tags are subjected to second-generation high-throughput sequencing, and the resulting data can be plotted to visualize the final bar-code counts for each DEL member. |
6. |
The eluted library members can be used for a further consecutive selection round against fresh, immobilized target before PCR amplification and sequencing. |

Analysis of DEL Data |
1. |
Decoding of DNA sequencing data using a data analysis program, and comparing with the corresponding DNA encoding and molecular building blocks during library design, will give the structure of the target compounds. |

Each axis of the cube graph provides the molecular building blocks used in the library preparation process, with each data point representing a specific enriched library member. |
