In antibody drug development and cellular trafficking research, it is essential to determine whether antibodies are truly internalized by cells. Traditional labeling methods often fail to distinguish surface-bound antibodies from those taken up into acidic intracellular compartments, leading to ambiguous results. Sino Biological is excited to launch a new product of SwiftStain™ human pH-red antibody labeling dye (Cat #: SSL001), a pH-sensitive fluorescent dye that activates only in low-pH organelles after internalization. It is an ideal tool for studying antibody internalization in applications such as ADC development and more.

Internalization assay using SwiftStain™ Human pH-red Antibody Labeling Dye (Cat#: SSL001)-labeled antibodies and antigen-expressing cells.
|
Her2+ SK-BR-3 cells (Fig. A) and CD33+ THP-1 cells (Fig. B) were incubated with Trastuzumab (IgG1) or Gemtuzumab (IgG4) conjugated to the dye, or with the Dye alone, at 37ºC, 5% CO2 for 17 hours, washed three times with 1x PBS, and then analyzed by flow cytometry.

Internalization assay using pH sensitive antibody labeling reagents from two vendors: a competitor and Sino Biological (Cat#: SSL001).
|
THP-1 (CD33+) cells (Fig. A) or SK-BR-3 (Her2+) cells (Fig. B) were treated with Gemtuzumab (Fig. A) or Trastuzumab (Fig. B) conjugated to the dyes from each vendor. Parallel cell samples that were treated with either antibody or dye alone served as negative controls.
